Amazon Plans Smartphone Comeback More Than a Decade After Fire Phone Flop (reuters.com) 46

Amazon is reportedly developing a new AI-focused smartphone that doesn't rely as heavily on traditional apps. "The phone is seen as a potential mobile personalization device that can sync with home voice assistant Alexa and serve as a conduit to Amazon customers throughout the day," reports Reuters. From the report: As envisioned, the new phone's personalization features would make buying from Amazon.com, watching Prime Video, listening to Prime Music or ordering food from partners like Grubhub easier than ever, the people said. They asked for anonymity because they were not authorized to discuss internal matters. A key focus of the Transformer project has been integrating artificial intelligence capabilities into the device, the people said. That could eliminate the need for traditional app stores, which require downloading and registering for applications before they can be used. Alexa would likely be a core feature but not necessarily the primary operating system of the phone, the people said. When Amazon launched the Fire Phone in 2014, it aimed to compete directly with offerings from Samsung and Apple. Instead, the device received mixed reviews and failed to impress reviewers, leading Amazon to abandon the effort just over a year later.

Plastic-Eating Bacteria Could Combat Pollution Problems, Scientists Hope (msn.com) 68

The Washington Post on scientists who "discovered that bacteria commonly found in wastewater can break down plastic to turn it into a food source, a finding that researchers hope could be a promising answer to combat one of Earth's major pollution problems." In a study published Thursday in Environmental Science and Technology, scientists laid out their examination of Comamonas testosteroni, a bacteria that grows on polyethylene terephthalate, or PET, a plastic commonly found in single-use food packaging and water bottles. PET makes up about 12 percent of global solid waste and 90 million tons of the plastic produced each year... Unlike most other bacteria, which thrive on sugar, C. testosteroni has a more refined palate, including chemically complex materials from plants and plastics that take longer to decompose.

The researchers are the first to demonstrate not only that this bacteria can break down plastic, but they also illuminate exactly how they do it. Through six meticulous steps, involving complex imaging and gene editing techniques, the authors found that the bacteria first physically break down plastic by chewing it into smaller pieces. Then, they release enzymes — components of a cell that speed up chemical reactions — to chemically break down the plastic into a carbon-rich food source known as terephthalate...

The bacteria take a few months to break down chunks of plastic, according to Rebecca Wilkes [a lead author on the study and postdoctoral researcher at the National Renewable Energy Laboratory]. As a result, if the bacteria are going to be efficient tools, a lot of optimization needs to take place to speed up the rate at which they decompose pollutants. One approach is to promote bacterial growth by providing them with an additional food source, such as a chemical known as acetate.

A senior author on the study (and associate professor of civil and environmental engineering at Northwestern University) tells the Washington Post that "The machinery in environmental microbes is still a largely untapped potential for uncovering sustainable solutions we can exploit."

Can Food Scientists Re-Invent Sugar? (msn.com) 102

The Wall Street Journal visits scientists at Harvard University's Wyss Institute for Biologically Inspired Engineering who are researching a "sugar-to-fiber" enzyme (normally used by plants to create stalks). They're testing a version they've "encased in spherical nanoparticles — tiny mesh-like cages made of pectin that allow the enzyme to be added to food without being activated until it reaches the intestine.

"Once there, a change in pH causes the cage to expand, freeing the enzyme to float through its holes and start converting sugar to fiber." The Wyss Institute's goal for its enzyme product was to reduce the sugar absorbed from food by 30%, though it has the potential to remove even more than that, says Sam Inverso, director of business development partnerships at the Wyss Institute. The enzyme's ability to turn sugar into fiber is also key, as most Americans don't get nearly enough fiber in their diet, says Adama Sesay, a senior engineer at the Wyss Institute who worked on the project...

The Wyss Institute is now licensing the technology to a company to help bring its enzyme product to market, a process that entails additional testing and work to secure regulatory approval. Inverso says that the aim is for the product to be available to U.S. food manufacturers within the next two years, and that other encapsulated enzymes could follow: products that reduce lactose absorption after drinking milk, or cut gluten after eating bread. For now the enzyme works better in solid food than in a liquid. Producing it in large quantities and at low cost is still a ways off — currently it's 100 times more expensive than raw sugar, Inverso says.

And the Journal notes they're not the only ones working on the problem: San Francisco-based startup Biolumen recently launched a product called Monch Monch, a drink mix made of fibrous, microscopic sponges designed to soak up sugar and prevent it from reaching the bloodstream. At mealtime consumers can blend a teaspoon of Monch Monch, which has no taste, smell or color, into drinks from water to wine. Once it has reached the stomach, the sponges start to swell and sequester sugar, reducing its burden on the body, says Dr. Robert Lustig, Biolumen's co-founder and chief medical officer... One gram of Monch Monch can sequester six grams of sugar, says Lustig... The product, introduced as a dietary supplement, can also be used as a food ingredient under a Food and Drug Administration principle known as "generally recognized as safe." Packets of Monch Monch are available for purchase online, and Biolumen says it is in talks with U.S. food manufacturers it declined to name about its use in other products...

Food companies are betting on other solutions for now. Cereal startup Magic Spoon uses allulose, a natural sugar found in figs and raisins that is growing in popularity, helped by FDA guidance that allows it to be excluded from sugar or added-sugar totals on nutrition labels. Ingredient company Tate & Lyle, which makes allulose from corn kernels, says the sweetener tastes like sugar and adds bulk and caramel color, but passes through the body without being metabolized... Chicago-based Blommer Chocolate recently launched a line of reduced-sugar chocolate and confectionery products made with Incredo, a sugar that has been physically altered to taste sweeter using a mineral carrier that dissolves faster in saliva and targets the sweet-taste receptors on the tongue. Incredo's use enables manufacturers to use up to 50% less sugar, the company says.

The article even notes that "researchers still working to reduce sugar are peddling new technologies, like individual sugar crystals modified to dissolve more quickly in the mouth, making food taste sweeter."

Vibrating Pill May Give Dieters a Feeling of Fullness, Study Suggests (theguardian.com) 56

Scientists have developed a vibrating pill that, when swallowed before eating, can create a feeling of fullness. The Guardian reports: The research, which has yet to be carried out in humans, shows that after 30 minutes of activity by the Vibes pill, pigs ate on average almost 40% less food in the following half hour than they did without the device, and gained weight more slowly. The Vibes name is an acronym derived from the pill's full title -- Vibrating Ingestible BioElectronic Stimulator. The work in pigs suggests the vibrations activate stretch receptors in the stomach, simulating the presence of food. This results in signals being sent to the hypothalamus in the brain via the vagus nerve, increasing levels of various hormones that give rise to a feeling of fullness and decreasing those that result in feelings of hunger.

"We envision the Vibes pill being ingested on a relatively empty stomach 20 to 30 min before anticipated meals to trigger the desired sensation of satiety early in the meal,â the team write, adding that when produced at scale, the cost of the pills is expected to be in the cents to one dollar range. The vibrations, which are powered by a battery encased in the swallowed capsule, can be triggered when stomach acid dissolves a membrane around the pill, or by a timer. The researchers say the pills, which are about the size of a large vitamin tablet, offer a non-invasive, temporary therapy, without the need for weight-loss surgery, and exit the body with other solid waste -- meaning in humans they are flushed down the toilet. However they suggest it could be possible to develop pills that are implanted, or stay in the stomach, to reduce the need for people to repeatedly take them, should they require continuing therapy.

Why Armored CAR T Therapy Is Our Best Shot At Curing Cancer (thedailybeast.com) 40

An anonymous reader quotes a report from Ars Technica: Immune therapies have rewritten the game when it comes to cancer treatment, earning the "fifth pillar" label next to more tried and true treatments like radiation therapy, surgery, and chemotherapy. And no immunotherapy has garnered quite the same excitement as CAR T-cell therapy, first approved in 2017 by the Food and Drug Administration to treat a form of acute lymphoblastic leukemia. At the time, then-FDA Commissioner Scott Gottlieb called the approval "a new frontier in medical innovation," and it seemed like the possibilities for CAR T were near-endless. Flash-forward almost six years, and six therapies have been approved for blood cancers, including lymphomas, leukemia, and multiple myeloma. There's no question that when CAR T works, it works incredibly well. But why it doesn't work for the majority of patients or cancer types has befuddled researchers.

CAR T therapies also haven't yet been expanded to treat solid tumors, which make up the majority of cancers. The immune therapy hasn't been able to crack physical barriers, idiosyncratic tumor cells, and a suppressive microenvironment that characterize these cancers. But a new generation of CAR T therapies are emerging, equipped with highly effective small molecules that scientists hope will solve their low success rate for both blood cancers and solid tumors. Known as "armored CAR T," these infusions have been boosted with additional layers of protection and cancer-fighting proteins. Early research shows that the armored flavor of CAR T might have what it takes for immunotherapy to go the extra mile.

Briefly, immunotherapy can boost or restore the body's immune system by lowering cancer cells' defenses, priming the immune system's T cells to destroy tumors, or -- in the case of CAR T-cell therapy -- genetically editing a patient's T cells. Scientists do this by isolating a patient's T cells from their blood and inserting a gene for a chimeric antigen receptor -- a type of synthetic protein that has been specially made to bind to another protein present on the surface of that patient's cancer cells. Then, upon infusing these modified T cells back into a patient, the immune fighters will recognize and destroy the tumor cells when the patient's normal T cells have failed. That, at least, is the idea. But when CAR T doesn't work, a few factors could be at play. One, Lim said, is the tumor microenvironment, the set of chemicals and structures present in solid cancers that naturally suppress pushback from the body's immune system. Tumor heterogeneity is also a factor -- depending on the type and stage of cancer, tumor cells may not express the protein that the CAR T cells' receptors have been designed to recognize, blocking the ability of the CAR T cells to attack the cancer Finally, there are physical barriers on the outside of a solid tumor that can even prevent the T cells from entering inside to destroy the cancer. Armored CAR T is meant to overcome these difficulties. With this form of therapy, not only are T cells engineered to express a tumor cell's surface protein, they are also given potent cargo often in the form of small proteins called cytokines. If deployed on their own, such molecules can be toxic -- but pairing them with T cells designed to release them at the tumor site and nowhere else represents a promising new strategy.
Jakub Svodoba, an oncologist at the University of Pennsylvania, "is helping to lead a clinical trial using armored CAR T-cell therapy to treat patients with non-Hodgkin lymphoma for whom previous CAR T therapy has failed," reports the Daily Beast. "Last month, he presented findings that the first seven patients treated with this therapy all responded to it and were alive eight months after receiving it. Svoboda said an important additional finding was that toxicities experienced by the patients -- a concern with cytokines -- were comparable to those from traditional CAR T therapy."

Wendell Lim, a cellular and molecular pharmacology researcher at the University of California, San Francisco, and his team are also working on an armored CAR-T therapy, with the potential for it to be used to treat solid tumors. "In a paper published in Science on Dec. 16, he and his colleagues designed T cells to release a cytokine directly to the tumors of mice with pancreatic cancer and melanoma," reports the Daily Beast. "In the study, they wrote that these cancers are 'nearly completely resistant' to treatment with traditional CAR T, but releasing a cytokine allowed the engineered T cells to get past the tumor microenvironment -- effectively solving one of the issues that has set the therapy back."

How Amazon Put Ukraine's 'Government in a Box' (latimes.com) 23

An anonymous reader shares a report: Since Februrary, Amazon has been playing Santa Claus to Ukraine, delivering planeloads of goods, including blankets, hygiene kits, diapers, food and toys, for the war-torn nation and refugees in Poland and other parts of Europe. But long term, what's more important to Ukrainians than the gifts coming in is what's going out: massive amounts of government, tax, banking and property data vulnerable to destruction and abuse should Russian invaders get their hands on it. Since the day Russia launched its invasion Feb. 24, Amazon has been working closely with the Ukrainian government to download essential data and ferry it out of the country in suitcase-sized solid-state computer storage units called Snowball Edge, then funneling the data into Amazon's cloud computing system.

"This is the most technologically advanced war in human history," said Mykhailo Fedorov, Ukraine's 31-year-old vice prime minister and minister of digital transformation, referring not just to weapons but data too. Amazon Web Services' "leadership made a decision that saved the Ukrainian government and economy." Amazon has invested $75 million so far in its Ukraine effort, which includes the data transfer via the Snowballs. Fedorov, speaking at a tech conference in Las Vegas this month, called it "priceless." The data, 10 million gigabytes so far, represent "critical information infrastructure. This is core for operation of the economy, of the tax system, of banks, and the government overall," he said. The data also include property records whose safekeeping can help prevent theft of Ukrainian homes, businesses and land.

Through history, invaders have "come in and staged fake referendum and parceled out the land to their chums," said Liam Maxwell, head of government transformation at Amazon Web Services, the company's highly profitable cloud computing arm. "That kind of thing has been happening since William the Conquerer." The Odessa Journal newspaper reported in June that residents of the Russian-occupied city of Mariupol whose homes had been destroyed were being moved into the homes of citizens who had fled the area, and were being forced to find those who left and pressure them to cooperate in some fashion with the Russians. Maxwell, who's based in London, had already been working with Ukraine for years when it became clear by January that Russia planned to attack the country.

Scientists Turn Plastic Into Diamonds In Breakthrough (vice.com) 46

An anonymous reader quotes a report from Motherboard: More than a billion miles away from Earth, on the ice giants of Neptune and Uranus, diamonds are forever. This isn't cosmic poetry, but a reasonable scientific conclusion: We know that under extreme pressures and high temperatures miles beneath a planet's surface, hydrocarbons are pummeled into a crystalline bling coveted by the affianced. But on far-flung Neptune and Uranus, the Universe's diamond-making process is a bit more curious. Since the 1970s, scientists believed that diamonds might actually rain down toward the mostly slushy planets' rocky interiors -- a diamond rain, if you will. In 2017, researchers in Germany and California found a way to replicate those planetary conditions, fabricating teeny tiny diamonds called nanodiamonds in the lab using polystyrene (aka Styrofoam). Five years later and they're back at it again, this time using some good ol' polyethylene terephthalate (PET), according to a study published on Friday in Science Advances. The research has implications not only for our understanding of space, but paves a path toward creating nanodiamonds that are used in a range of contexts out of waste plastic.

So, why in the world are we making diamonds out of the same plastic that things like food containers and water bottles are made of? There's a good reason for this, Dominik Kraus, a scientist at the German research laboratory Helmholtz-Zentrum Dresden-Rossendorf and lead author of the study, said in an email. When Kraus and his colleagues first attempted making nanodiamonds with polystyrene -- which contains the same elements of carbon and hydrogen found on Neptune and Uranus -- they did so by bombarding the material with the Linac Coherent Light Source, a high-powered X-ray laser at the SLAC National Acceleratory Laboratory in California. This process rapidly heated the polystyrene to 5,000 Kelvin (around 8,540 degrees Fahrenheit) and compressed it by 150 gigapascals, similar to conditions found about 6,000 miles into the interior of the icy planets. While the researchers were able to make the microscopic bling with two quick hits from the laser, they later realized one vital chemical ingredient was missing: oxygen. So they turned to PET, which has a good balance of not only carbon and hydrogen but also oxygen, making it a closer chemical proxy to the ice giants than polystyrene.

"The chemistry at these conditions is very complex and modeling extremely difficult. 'Anything can happen' is a typical phrase when discussing such scenarios with theorists," said Kraus. "Indeed, there were some predictions showing that the presence of oxygen is helping [carbon separate from hydrogen] and diamond formation, but also ideas that it may be the other way around." To put the theoretical pedal to the metal, Kraus and his colleagues took a piece of PET, put it through the same 2017 experimental motions, but also included something called small angle X-ray diffraction to see how quickly and how large the diamonds grow. "We found that the presence of oxygen enhances diamond formation instead of preventing it, making 'diamond rain' inside those planets a more likely scenario," said Kraus. "We [also] see that diamonds grow larger for higher pressures and with progressing time in the experiments." They were also able to squeeze out a lot of tiny diamonds from just one shot of X-ray, on the order of a few billion crystallites (or a few micrograms if you're talking total weight). But Kraus said this isn't enough, at least right now, for application purposes like diamond quantum sensors, which are used to detect magnetic flow, or chemical catalysts, which need a couple of milligrams at minimum. However, it could eventually be scaled up to serve those purposes, and be the first step to a more ritzy way of plastic recycling.
Kraus and his team also believe they found more evidence for superionic water, a bizarre type of water that acts like a weird cross between solid and liquid. "Kraus said that the finding that nanodiamonds indeed form inside ice giants makes it more likely for the conditions for superionic water to arise," reports Motherboard.

Kraus said: "[O]ur experiments show that carbon is separating from hydrogen and oxygen allowing pure water regions to form inside the planets. Thus, by making diamond precipitation a more realistic scenario inside those planets, also the formation of superionic water becomes more likely."

Why Bill Gates Supports America's Climate Legislation (nytimes.com) 128

Bill Gates wrote a New York Times opinion piece Saturday arguing that it's immediately clear we're experiencing the effects of climate change. Just in America, "Extreme heat and drought are affecting tens of millions of people, as floods and wildfires ravage communities from Appalachia to California."

So he's urging the U.S. Congress to "face down the climate crisis" while "strengthening our country's energy security, creating opportunities for businesses and improving the lives of Americans," by supporting a massive climate-spending bill being voted on this weekend in the U.S. Senate. Gates calls the bill "our best chance to build an energy future that is cleaner, cheaper and more secure." Many of the technologies we'll need to reach net-zero emissions don't exist, are in early stages of development or are still too expensive to scale up. At the same time, more mature technologies like solar, wind and electric vehicles must be deployed more quickly in more places. Through new and expanded tax credits and a long-term approach, this bill would ensure that critical climate solutions have sustained support to develop into new industries....

With those incentives and investments, this bill would catalyze a new era of American innovation. The ability of America's universities and industries to innovate remains second to none, yet the country risks falling behind as other countries race to build their own clean energy economies. This legislation would help turn American energy innovations into American energy industries and unlock huge economic opportunities in the energy market. If it becomes law, few nations would have the capacity for producing homegrown clean energy like the United States. America could quickly become a leader in the deployment of clean energy at the scale required.

Solving climate change is perhaps the hardest challenge humanity has ever faced. It will require fundamentally transforming how we power our communities, move goods, build things, heat and cool buildings and grow food — basically how we do everything. We need to do it rapidly with a cohesive and coherent plan if we want to avoid the worst effects of climate change.

The country has an opportunity to set an example by offering a vision of what's possible — and then by making it happen.

Gates also writes that the legislation "would begin to transform the parts of our economy that are hardest to decarbonize, like manufacturing, which we must do to reach net-zero emissions...

"Businesses are waiting on the sidelines for a strong signal from government that clean industries are a solid long-term investment. Passing the Inflation Reduction Act would send that message and enable private capital to supercharge our clean energy future with even greater confidence."

Crypto Enthusiasts' DAO Tries Raising $4 Billion to Buy a Football Team (cnbc.com) 48

In NFL news, an American football team — the Denver Broncos — "are up for sale," reports CNBC, "and a group of crypto enthusiasts is aiming to raise more than $4 billion using a decentralized autonomous organization, or DAO, to make it their own...." This group includes an eclectic mix of attorneys, accountants, software developers, pro athletes, and at least one mathematician. One of the people spearheading the cause is Sean O'Brien, who spent over a decade in Cisco's legal department, before leaving the corporate world to run a few small businesses with his wife. "We know it sounds a bit crazy, but it's also a bit badass," said O'Brien. "The purpose essentially is to establish an infrastructure so that fans from all walks of life can be owners of the Denver Broncos."

The pro football team is valued at just under $4 billion, and it is expected to garner the biggest price tag in North American sports history, according to ESPN....

DAOs take coordination of resources on the internet to a new level, according to Auston Bunsen, co-founder of QuikNode, which provides blockchain infrastructure to developers and companies. "They represent a new kind of organization moving at hyper speed," said Bunsen. Investor Cooper Turley, who has helped build several popular DAOs, says they're like an "internet community with a shared bank account."

"Basically, a small group of people come together to form a chat group, and then they decide to pull capital together, [typically] using an Ethereum wallet," Turley previously told CNBC....

Though the group will still fundraise in cryptocurrency, the idea is to give people partial ownership, in which they will participate in deciding how the team is run... The BuyTheBroncos group also has a pretty solid plan B. Organizers tell CNBC the more realistic goal is to raise around 25% of the money needed to place the winning bid, and from there, join forces with a consortium of more traditional buyers to make up the difference.

The group's Twitter account had less than 50 followers before CNBC's article — but 390 followers after the article ran. And O'Brien tells CBNC his ultimate goal is "to essentially open up peoples' eyes to what a DAO can do in the real world and make a tangible connection between this web3 life and the real world.

"Our thought is that it accelerates DAO adoption for solving real-world problems such as food scarcity or unhoused peoples."

Spinach Gives Fuel Cells a Power Up (ieee.org) 17

Researchers at the Department of Chemistry, American University, used spinach to make a carbon-rich catalyst that can be used to improve the performance of fuel cells and metal-air batteries. IEEE Spectrum reports: The spinach was a used a precursor for high-performance catalysts required for the oxygen reduction reactions (ORRs) in fuel cells. Traditionally, fuel cells have used platinum-based catalysts, but not only is platinum very expensive and difficult to obtain, it can be vulnerable to chemical poisoning in certain conditions. Consequently, researcher have looked into biomass-derived, carbon-based, catalysts to replace platinum, but there have been bottlenecks in preparing the materials in a cost-effective and non-toxic way. "We were a little bit lucky to pick up spinach," says [Shouzhong Zou], because of its high iron and nitrogen content. "At this point [our method] does require us to add a little bit more nitrogen into the starting material, because even though [spinach] has a lot of nitrogen to begin with, during the preparation process, some of this nitrogen gets lost."

The preparation of the spinach-based catalyst sounds as first suspiciously like a smoothie recipe at first -- wash fresh leaves, pulverize into a juice, and freeze-dry. This freeze-dried juice is then ground into a powder, to which melamine is added as a nitrogen promoter. Salts like sodium chloride and potassium chloride -- "pretty much like the table salt that we use in our kitchen," says Zou -- are also added, necessary for creating pores that increase the surface area available for reactions. Nanosheets are produced from the spinach -- melamine -- salt composites by pyrolyzing them at 900 C a couple of times. "Obviously... we can optimize how we prepare this material [to make it more efficient]."

An efficient catalyst means a faster, more efficient reaction. In the case of fuel cells, this can increase the energy output of batteries. This is where the porosity of the nanosheets helps. "Even though we call them nanosheets," Zou says, "when they are stacked together, it's not like a stack of paper that is very solid." The addition of salts to create tiny holes that allows oxygen to penetrate the material rather than access only the outer surfaces. "We need to make it porous enough that... all the active sites can be used." The other factor that favorably disposed the American University team towards spinach was that it is a renewable source of biomass. "Sustainability is a very important factor in our consideration," says Zou. The big question to explore, he adds, is how can we avoid competition "with the dinner table." (Biofuel production has already raised concerns about food crops being diverted away from hungry mouths.) "And the second is, how do we keep the carbon footprint down in terms of his catalyst preparation... because currently we do use high temperatures in our preparation procedure?" If we can find different ways to do these to achieve the same type of material, that will cut back the energy consumption and reduce significantly the carbon footprint."

A New Use For McDonald's Used Cooking Oil: 3D Printing (cnn.com) 73

An anonymous reader quotes a report from CNN: Professor Andre Simpson had a problem. The University of Toronto's Scarborough campus was paying through the nose for a crucial material for its 3D printer. Few would have guessed McDonald's would come to the rescue. Simpson is director of the school's Environmental NMR Center dedicated to environmental research. Central to this research is an analytical tool called the NMR spectrometer. NMR stands for nuclear magnetic resonance and is technically similar to how an MRI works for medical diagnostics. Simpson had bought a 3D printer for the lab in 2017. He hoped to use it to build custom parts that kept organisms alive inside of the NMR spectrometer for his research. But the commercial resin he needed for high-quality light projection 3D printing (where light is used to form a solid) of those parts was expensive.

The dominant material for light projection printing is liquid plastic, which can cost upward of $500 a liter, according to Simpson. Simpson closely analyzed the resin and spotted a connection. The molecules making up the commercial plastic resin were similar to fats found in ordinary cooking oil. What came next was the hardest part of the two-year experiment for Simpson and his team of 10 students -- getting a large sample batch of used cooking oil. "We reached out to all of the fast-food restaurants around us. They all said no," said Simpson. Except for McDonald's.
After filtering out chunks of food particles and experimenting with the oil for several months, the team was able to successfully print a high-quality butterfly with details as minute as 100 micrometers in size.

Psilocybin For Major Depression Granted Breakthrough Therapy By FDA 89

An anonymous reader quotes a report from New Atlas: The US Food and Drug Administration (FDA) has granted psilocybin therapy a Breakthrough Therapy designation for the second time in a year, this time with a view on accelerating trials testing its efficacy treating major depressive disorder (MDD). This new FDA Breakthrough Therapy approval focuses on a seven-site, Phase 2 trial currently underway in the United States. Coordinated by a non-profit research organization called the Usona Institute, the trial is exploring the antidepressant properties of a single psilocybin dose in treating patients with major depressive disorder.

Last year's Breakthrough Therapy designation was targeted at the drug's efficacy for treatment-resistant depression (TRD). That particular clinical classification categorizes patients suffering from MDD who have not responded effectively to at least two different pharmacological antidepressant treatments during a current depressive episode. It is estimated between 10 and 30 percent of MDD patients fall into the category of TRD. The new FDA approval focuses on Usona's research into the broader condition of MDD, characterized by episodes of severe depression that last more than two weeks. Hundreds of millions of people around the world suffer from these kinds of acute major depressive episodes every year.
"The Usona Phase 2 trial plans to enrol 80 subjects, randomized to receive either a single active dose of psilocybin or an active placebo containing niacin," the report adds. "The methodology being trialed is similar to other psilocybin therapy studies, with a number of preparatory psychotherapy sessions preceding the active psychedelic dose, and a number of integrative psychotherapy sessions afterwards." It's estimated that the current Phase 2 trial will be completed by early 2021, where it should be able to move into larger Phase 3 trials if the results are positive.

The FDA also gave psilocybin therapy a Breakthrough Therapy status late last year. These trials should be completed sometime in 2020, "suggesting the next 12 to 24 months will offer some compelling and solid data into how effective this new psychedelic therapy actually is in treating several different forms of depression," reports New Atlas.

Making Buildings, Cars and Planes From Materials Based on Plant Fibres (economist.com) 51

Materials-science researchers are finding that plant fibres can add durability and strength to substances already used in the construction of buildings and in goods that range from toys and furniture to cars and aircraft. From a report: A big bonus is that, because plants lock up carbon in their structure, using their fibres to make things should mean less carbon dioxide is emitted. The production of concrete alone represents some 5% of man-made global CO{-2} emissions, and making 1kg of plastic from oil produces 6kg of the greenhouse gas. Start with the carrots. These are being investigated by Mohamed Saafi at Lancaster University, in England. Dr Saafi and his colleagues do not use whole carrots, but rather what they call "nanoplatelets" that have been extracted from carrots discarded by supermarkets or as waste from food-processing factories. Sugar-beet peelings are also a useful source of nanoplatelets.

The researchers are working with CelluComp, a British firm that produces such platelets for industrial applications, including as an additive that helps toughen the surface of paint as it dries. Each platelet is only a few millionths of a metre across. It consists of a sheet of stiff cellulose fibres. Although the fibres are minute, they are strong. By combining platelets with other materials a powerful composite can be produced. Dr Saafi is mixing the platelets into cement, which is made by burning limestone and clay together at high temperature. To turn cement into concrete it is mixed with aggregates such as sand, stones and crushed rocks, which act as reinforcement, and with water, which reacts with the chemicals in the cement to form a substance called calcium silicate hydrate. This starts off as a thick gel, but then hardens into a solid matrix that binds the aggregates together.

Microbes May Help Astronauts Transform Human Waste Into Food (phys.org) 103

A Penn State researcher team has shown that it is possible to rapidly break down solid and liquid waste to grow food with a series of microbial reactors, while simultaneously minimizing pathogen growth. They reported their findings in the journal Life Sciences in Space Research. Phys.Org reports: To test their idea, the researchers used an artificial solid and liquid waste that's commonly used in waste management tests. They created an enclosed, cylindrical system, four feet long by four inches in diameter, in which select microbes came into contact with the waste. The microbes broke down waste using anaerobic digestion, a process similar to the way humans digest food. The team found that methane was readily produced during anaerobic digestion of human waste and could be used to grow a different microbe, Methylococcus capsulatus, which is used as animal feed today. The team concluded that such microbial growth could be used to produce a nutritious food for deep space flight. They reported in Life Sciences in Space Research that they grew M. capsulatus that was 52 percent protein and 36 percent fats, making it a potential source of nutrition for astronauts.

Because pathogens are also a concern with growing microbes in an enclosed, humid space, the team studied ways to grow microbes in either an alkaline environment or a high-heat environment. They raised the system's pH to 11 and were surprised to find a strain of the bacteria Halomonas desiderata that could thrive. The team found this bacteria to be 15 percent protein and 7 percent fats. At 158 degrees Fahrenheit, which kills most pathogens, they grew the edible Thermus aquaticus, which consisted of 61 percent protein and 16 percent fats.

'Watershed' Medical Trial Proves Type 2 Diabetes Can Be Reversed (bbc.com) 224

dryriver writes: For those suffering from type 2 diabetes, there is good news. Nearly half of the participants in a watershed trial of a new diabetes treatment were able to reverse their affliction. The method is quite simple -- an all liquid diet that causes participants to lose a lot of weight, followed by a carefully controlled diet of real solid foods. Four times a day, a sachet of powder is stirred in water to make a soup or shake. They contain about 200 calories, but also the right balance of nutrients. If the patient can keep away from other foods long enough, there is a chance of reversing type 2 diabetes completely. Prof Roy Taylor, from Newcastle University, told the BBC: "It's a real watershed moment. Before we started this line of work, doctors and specialists regarded type 2 as irreversible. But if we grasp the nettle and get people out of their dangerous state (being overweight), they can get remission of diabetes." However, doctors are not calling this a cure. If the weight goes back on, then the diabetes will return. The trial only looked at people diagnosed with diabetes in the last six years. Doctors believe -- but do not know with absolute certainty yet -- that in people who have had the affliction much longer than that, there may be too much permanent damage to make remission possible. The trial results have been published in the Lancet medical journal.

Artificial Sweeteners Associated With Weight Gain, Heart Problems In Analysis of Data From 37 Studies (npr.org) 374

In practice, it's not so simple, as a review of the scientific evidence on non-nutritive sweeteners published Monday shows. After looking at two types of scientific research, the authors conclude that there is no solid evidence that sweeteners like aspartame and sucralose help people manage their weight. And observational data suggest that the people who regularly consume these sweeteners are also more likely to develop future health problems, though those studies can't say those problems are caused by the sweeteners.

The review, published Monday in the Canadian Medical Association Journal, looked at 37 studies. Seven of them were randomized trials, covering about 1,000 people, and the rest were observational studies that tracked the health and habits of almost 406,000 people over time.

They have developed a coating that makes bottle interiors super slippery. The coating can also be used to make it easier to squeeze out the contents of other containers, such as those holding toothpaste, cosmetics and even glue. The researchers at the Massachusetts Institute of Technology (MIT) believe that their innovation could dramatically reduce waste. In its manufacture, the container must first be coated on the inside with a rough surface. A very thin layer is then placed over this. And, finally, a liquid is added that fills in any troughs to form a very slippery surface -- like an oily floor. The ketchup hovers on top and just glides out of the bottle. According to Prof Kripa Varanasi, who developed the slippery surface, the technology is completely safe. "The cool thing about it is that because the coating is a composite of solid and liquid, it can be tailored to the product. So for food, we make the coating out of food-based materials and so you can actually eat it."

A recent survey of over 2,000 adults conducted by Harris Poll on behalf of Dashlane, a "leader in online identity and password management," found that nearly 40 percent of Americans would give up sex for an entire year if it meant they'd never have to worry about being hacked. Huffington Post reports: 40 percent of people also said they'd give up their favorite food for one month in the name of peace of mind online. If all of this sounds drastic, the truth is that it probably is. The single biggest thing people can do to help keep their online identity safe is probably the easiest -- a solid password. 10 years ago, anti-virus was the primary method of online security. But since the Internet has left the desktop and is on laptops, tablets, and cell phones, and since so many people now use the cloud for backing up their sensitive data, following proper password protocol is critical. Of course, having a solid password doesn't do a lot of good if you're giving it out to people. And nearly 50% of people have shared a password to an e-mail account or to an account like Netflix with a friend or had a friend share theirs (which is a surprisingly high number when you consider that 4 out of 10 people said that sharing an online social media password was more intimate than sex). A look at the password habits of Americans showed that about 30% have used a pet's name, almost 25% have used a family member's name, 21% a birthday, and 10% each have used an anniversary, a sports team, an address, or a phone number. So if you just know a few basic, personal details about someone, you've got a decent chance at cracking their password. The study also revealed some interesting data in that younger Americans (those age 18 to 34) who grew up online are far more trusting with passwords than older generations, and married people are less likely to part with passwords than single people.

But unfortunately Mars mania reflects an excessively optimistic view of what it actually takes to travel to and live on Mars, papering over many of the harsh realities and bitter truths that underlie the dream. "First, there is the tedious business of getting there. Using current technology and conventional chemical rockets, a trip to Mars would be a grueling, eight- to nine-month-long nightmare for the crew," writes Regis. "Tears, sweat, urine and perhaps even solid waste will be recycled, your personal space is reduced to the size of an SUV., and you and your crewmates are floating around sideways, upside down and at other nauseating angles." According to Regis every source of interpersonal conflict, and emotional and psychological stress that we experience in ordinary, day-to-day life on Earth will be magnified exponentially by restriction to a tiny, hermetically sealed, pressure-cooker capsule hurtling through deep space and to top it off, despite these constraints, the crew must operate within an exceptionally slim margin of error with continuous threats of equipment failures, computer malfunctions, power interruptions and software glitches.

But getting there is the easy part says Regis. "Mars is a dead, cold, barren planet on which no living thing is known to have evolved, and which harbors no breathable air or oxygen, no liquid water and no sources of food, nor conditions favorable for producing any. For these and other reasons it would be accurate to call Mars a veritable hell for living things, were it not for the fact that the planet's average surface temperature is minus 81 degrees Fahrenheit." These are only a few of the many serious challenges that must be overcome before anyone can put human beings on Mars and expect them to live for more than five minutes says Regis. "The notion that we can start colonizing Mars within the next 10 years or so is an overoptimistic, delusory idea that falls just short of being a joke."
