Rare Genetic Disorder Treated in Womb For the First Time (nature.com) 30

A two-and-a-half-year-old girl shows no signs of a rare genetic disorder, after becoming the first person to be treated for the motor-neuron condition while in the womb. Nature: The child's mother took the gene-targeting drug during late pregnancy, and the child continues to take it. The "baby has been effectively treated, with no manifestations of the condition," says Michelle Farrar, a paediatric neurologist at UNSW Sydney in Australia. The results were published in the New England Journal of Medicine yesterday. The child was conceived with a genetic condition known as spinal muscular atrophy, which affects motor neurons that control movement, and leads to progressive muscle weakening.

About one in every 10,000 births have some form of the condition -- making it a leading genetic cause of death in infants and children. In its most severe form, as in the case of this child, individuals lack both copies of the SMN1 gene, and have only one or two copies of a neighbouring gene, SMN2, that partially compensates for that deficiency. As a result, the body does not produce enough of the protein required for maintaining motor neurons in the spinal cord and brainstem. This protein is most important in the second and third trimesters, and the first few months of life. Babies with severe disease don't usually live past their third birthday.

Men Have Grown Twice As Much As Women Over Past Century, Study Shows 167

According to a new study published in the journal Biology Letters, men around the world have gained height and weight twice as fast as women over the past century. The Guardian reports: "We're seeing insights into how sexual selection has shaped the male and female body and how improved environments, in terms of food and a lower burden of disease, have freed us from our shackles," said Prof Lewis Halsey at the University of Roehampton. Halsey and his colleagues used data from the World Health Organization, overseas authorities and UK records to see how height and weight have changed with living conditions. The latter was measured by the human development index (HDI), a score based on life expectancy, time in education and per capita income, which ranges from zero to one.

Analysis of records from dozens of countries found that for every 0.2 point increase in HDI, women were on average 1.7cm taller and 2.7kg heavier, while men were 4cm taller and 6.5kg heavier. This suggests that as living conditions improve, both height and weight increase, but more than twice as fast in men than women. To see whether similar trends played out within countries, the researchers delved into historical height records in the UK where HDI rose from 0.8 in 1900 to 0.94 in 2022. During the first half of the century, average female height increased 1.9% from 159cm to 162cm, while average male height rose 4% from 170cm to 177cm. "To put this in perspective, about one in four women born in 1905 was taller than the average man born in 1905, but this dropped to about one in eight women for those born in 1958," Halsey said.

Writing in Biology Letters in a study titled "The sexy and formidable male body: men's height and weight are condition-dependent, sexually selected traits," the scientists speculate that women's sexual preferences may have fueled a trend for taller, more muscular men -- although in an age of obesity, heavy does not necessarily mean muscular. Stature and physique are prime indicators of health and vitality, Halsey said, while sexual selection also favors men who are better able to protect and defend their partners and offspring against others. "Women can find men's height attractive because, potentially, it makes them more formidable, but also because being taller suggests they are well-made," said Halsey. "As they've grown up, they haven't been affected by the slings and arrows of a bad environment, so they've reached more of their height potential. It's an indicator that they're well-made."

US Fines T-Mobile $60 Million, Its Largest Penalty Ever, Over Unauthorized Data Access (reuters.com) 12

The U.S. Committee on Foreign Investment (CFIUS) fined T-Mobile $60 million, its largest penalty ever, for failing to prevent and report unauthorized access to sensitive data tied to violations of a mitigation agreement from its 2020 merger with Sprint. "The size of the fine, and CFIUS's unprecedented decision to make it public, show the committee is taking a more muscular approach to enforcement as it seeks to deter future violations," reports Reuters. From the report: T-Mobile said in a statement that it experienced technical issues during its post-merger integration with Sprint that affected "information shared from a small number of law enforcement information requests." It stressed that the data never left the law enforcement community, was reported "in a timely manner" and was "quickly addressed." The failure of T-Mobile to report the incidents promptly delayed CFIUS' efforts to investigate and mitigate any potential harm to U.S. national security, they added, without providing further details. "The $60 million penalty announcement highlights the committee's commitment to ramping up CFIUS enforcement by holding companies accountable when they fail to comply with their obligations," one of the U.S. officials said, adding that transparency around enforcement actions incentivizes other companies to comply with their obligations.

Chinese Drone Maker DJI Might Get Banned Next in the US (nytimes.com) 107

U.S. authorities consider DJI a security threat. Congress is weighing legislation to ban it [non-paywalled link], prompting a lobbying campaign from the company, which dominates the commercial and consumer drone markets. The New York Times: DJI is on a Defense Department list of Chinese military companies whose products the U.S. armed forces will be prohibited from purchasing in the future. As part of the defense budget that Congress passed for this year, other federal agencies and programs are likely to be prohibited from purchasing DJI drones as well. The drones -- though not designed or authorized for combat use -- have also become ubiquitous in Russia's war against Ukraine.

The Treasury and Commerce Departments have penalized DJI over the use of its drones for spying on Uyghur Muslims who are held in camps by Chinese officials in the Xinjiang region. Researchers have found that Beijing could potentially exploit vulnerabilities in an app that controls the drone to gain access to large amounts of personal information, although a U.S. official said there are currently no known vulnerabilities that have not been patched. Now Congress is weighing legislation that could kill much of DJI's commercial business in the United States by putting it on a Federal Communications Commission roster blocking it from running on the country's communications infrastructure.

The bill, which has bipartisan support, has been met with a muscular lobbying campaign by DJI. The company is hoping that Americans like Mr. Nordfors who use its products will help persuade lawmakers that the United States has nothing to fear -- and much to gain -- by keeping DJI drones flying. "DJI presents an unacceptable national security risk, and it is past time that drones made by Communist China are removed from America," Representative Elise Stefanik, Republican of New York and one of the bill's primary sponsors, said in an emailed statement this month.

'Cory Doctorow Has a Plan To Wipe Away the Enshittification of Tech' (theregister.com) 206

In an interview with The Register, author and activist Cory Doctorow offers potential solutions to stop "enshittification," an age-old phenomenon that has become endemic in the tech industry. It's when a platform that was once highly regarded and user-friendly gradually deteriorates in quality, becoming less appealing and more monetized over time. Then, it dies. Here's an excerpt from the interview, conducted by The Register's Iain Thomson: [...] Doctorow explained that the reasons for enshittification are complex, and not necessarily directly malicious -- but a product of the current business environment and the state of regulation. He thinks the way to flush enshittification is enforcing effective competition. "We need to have prohibition and regulation that prohibits the capital markets from funding predatory pricing," he explained. "It's very hard to enter the market when people are selling things below cost. We need to prohibit predatory acquisitions. Look at Facebook: buying Instagram, and Mark Zuckerberg sending an email saying we're buying Instagram because people don't like Facebook and they're moving to Instagram, and we just don't want them to have anywhere else to go."

The frustrating part of this is that the laws needed to break up the big tech monopolies that allow enshittification, and encourage competition, are already on the books. Doctorow lamented those laws haven't been enforced. In the US, the Clayton Act, the Federal Trade Act, and the Sherman Act are all valid, but have either not been enforced or are being questioned in the courts. However, in the last few years that appears to be changing. Recent actions by increasingly muscular regulatory agencies like the FTC and FCC are starting to move against the big tech monopolies, as well as in other industry sectors. What's more, Doctorow pointed out, these are not just springing from the Democratic administration but are being actively supported by an increasing number of Republicans. He cited Lina Khan, appointed as chair of the FTC in part thanks to the support of Republican politicians seeking change (although the GOP now regularly criticizes her positions).

The sheer size of the largest tech companies certainly gives them an advantage in cases like these, Doctorow opined, noting that we've seen this in action more than 20 years ago. "Think back to the Napster era, and compare tech and entertainment. Entertainment was very concentrated into about seven big firms and they had total unity and message discipline," Doctorow recalled. "Tech was a couple of hundred firms, and they were much larger -- like an order of magnitude larger in aggregate than entertainment. But their messages were all over the place, and they were contradicting each other. And so they just lost, and they lost very badly."
Doctorow discusses the detrimental effects of mega-companies on innovation and security, noting how growth strategies focused on raising costs and reducing value can lead to vulnerabilities and employee demoralization. "Remember when tech workers dreamed of working for a big company before striking out on their own to put that big company out of business? Then that dream shrank to working for a few years, quitting and doing a fake startup to get hired back by your old boss in the world's most inefficient way to get a raise," he told the Def Con crowd last August. "Next it shrank even further. You're working for a tech giant your whole life but you get free kombucha and massages. And now that dream is over and all that's left is work with a tech giant until they fire your ass -- like those 12,000 Googlers who got fired six months after a stock buyback that would have paid their salaries for the next 27 years. We deserve better than this."

Additionally, Doctorow emphasizes the growing movement toward labor organizing in the tech industry, which could be a pivotal factor in reversing the trend of enshittification. "We're so much closer to tech unionization than we were just a few years ago. Yeah, it's still nascent, and yes, it's easy to double small numbers, but the strength is doubling very quickly and in a very heartening way," Doctorow told The Register. "We're really at a turning point. And some of it is coming from the kind of solidarity like you see with warehouse workers and tech workers."

Ultimately, Doctorow argues it should be possible to reintroduce a more competitive and innovative tech industry environment, where the interests of users, employees, and investors are better balanced.

The Internal Revenue Service has started using artificial intelligence to investigate tax evasion at multibillion-dollar partnerships as it looks for ways to better police hedge funds, private equity groups, real estate investors and large law firms. From a report: The announcement on Friday demonstrated how a more muscular I.R.S. is using some of the $80 billion allocated through last year's Inflation Reduction Act to target the wealthiest Americans and tackle the kinds of cases that had become too complex and cumbersome for the beleaguered agency to handle. The agency's new funding is intended to help the I.R.S. raise more federal revenue by cracking down on tax cheats and others who use sophisticated accounting maneuvers to avoid paying what they owe. But the allocation has been politically contentious, with Republicans claiming that the I.R.S. will use the funding to harass small businesses and middle-class taxpayers.

Earlier this year, Republicans succeeded in clawing back $20 billion as part of an agreement to raise the nation's borrowing cap. That political fight has put the onus on Democrats and the Biden administration to show that the funding is primarily enabling the I.R.S. to target the rich. "These are complex cases for I.R.S. teams to unpack," Daniel Werfel, the I.R.S. commissioner, said in a briefing with reporters. US Judge Temporarily Blocks Microsoft Acquisition of Activision (reuters.com) 40

A U.S. judge has granted the FTC request to temporarily block Microsoft's acquisition of Activision Blizzard, scheduling a hearing for a preliminary injunction and preventing the deal from closing until a court ruling is made. Reuters reports: U.S. District Judge Edward Davila scheduled a two-day evidentiary hearing on the FTC's request for a preliminary injunction for June 22-23 in San Francisco. Without a court order, Microsoft could have closed on the $69 billion deal as early as Friday. Davila said the temporary restraining order "is necessary to maintain the status quo while the complaint is pending (and) preserve this court's ability to order effective relief in the event it determines a preliminary injunction is warranted and preserve the FTC's ability to obtain an effective permanent remedy in the event that it prevails in its pending administrative proceeding."

Microsoft and Activision must submit legal arguments opposing a preliminary injunction by June 16; the FTC must reply on June 20. Davila said the bar on closing will remain in place until at least five days after the court rules on the preliminary injunction request. The case reflects the muscular approach to antitrust enforcement taken by the administration of U.S. President Joe Biden.

Universal Product Code Barcode Will Be Supplanted By 2027 With a More Data-Rich '2D' Barcode (axios.com) 206

The humble and familiar barcode -- a staple on consumer packaging for nearly 50 years -- will soon be replaced with a more robust and muscular successor that offers far more information about the product inside. Axios reports: In a worldwide push called "Sunrise 2027," the retail industry is transitioning from the standard 12-digit barcode -- that square of vertical lines that's printed on a package and makes it go "beep" at the checkout scanner -- to a two-dimensional web-enabled version. The effort is being orchestrated by GS1 US, the nonprofit standards organization that oversees the barcode world. In the United States, Universal Product Code (UPC) barcodes will be supplanted by a new 2D type, with information encoded on both the horizontal and vertical axes. By 2027, only the 2D barcodes will be accepted at registers globally.

The new "2D" barcodes will unlock reams of online extras (for consumers) and revolutionize inventory management (for retailers). Scanning them may tell us the field where something was grown, the factory where a garment was sewn, the sustainability practices of the company that made it -- or the washing instructions. [...] Stores will be able to respond immediately to product recalls, identifying faulty items and removing them from shelves. They'll be able to flag foods that are approaching their sell-by date -- and offer discounts before they expire. Consumers will gain online access to a trove of useful data -- everything from ingredients, recipes and potential allergens to promotional offers and information about how to recycle the product.

GS1 US just released a "barcode capabilities test kit" to help retailers evaluate their readiness for the 2D transition. We can expect to start seeing more products printed with 2D barcodes (or both types, as the transition moves forward) fairly soon.

Ministers must consider changing the law to allow scientists to carry out genome editing of human embryos for serious genetic conditions -- as a matter of urgency. That is the key message of a newly published report by a UK citizens' jury made up of individuals affected by genetic conditions. From a report: The report is the first in-depth study of the views of individuals who live with genetic conditions about the editing of human embryos to treat hereditary disorders and will be presented at the Third International Summit on Human Genome Editing, which opens at the Crick Institute in London this week. Scientists say that in a few years, they will be ready to use genome editing techniques to alter genes and induce changes in physical traits, such as disease risk, in future generations. In the UK, around 2.4 million people live with a genetic condition. These include cystic fibrosis, sickle cell disease, muscular dystrophy, various cancers, and some forms of hereditary blindness.

"Genome editing offers the prospect of preventing such conditions affecting future generations but there needs to be a full national debate on the issues," said Prof Anna Middleton of Cambridge University, the project's leader. "These discussions need to start now because genome editing is advancing so quickly. Many affected individuals want to debate the ethical issues and explore what implementation might look like." Genome editing acts like a pair of molecular scissors that can cut a strand of DNA at a specific site, allowing scientists to alter the structure of a gene, a form of manipulation that does not involve the introduction of DNA from other organisms. In the UK, as in most countries worldwide, it is illegal to perform genome editing on embryos that lead to pregnancy.

FDA Approves Most Expensive Drug Ever, a $3.5 Million-per-Dose Gene Therapy For Hemophilia B (cbsnews.com) 195

U.S. health regulators this week approved the first gene therapy for hemophilia, a $3.5 million one-time treatment for the blood-clotting disorder. From a report: The Food and Drug Administration cleared Hemgenix, an IV treatment for adults with hemophilia B, the less common form of the genetic disorder which primarily affects men. Currently, patients receive frequent, expensive IVs of a protein that helps blood clot and prevent bleeding. Drugmaker CSL Behring, based in Pennsylvania, announced the $3.5 million price tag shortly after the FDA approval, saying its drug would ultimately reduce health care costs because patients would have fewer bleeding incidents and need fewer clotting treatments.

According to a study cited by the National Library of Medicine, the price makes Hemgenix the most expensive medicine in the world, easily topping Novartis' Zolgensma gene therapy for spinal muscular atrophy (SMA), which costs right around $2 million per dose and is also a single-dose medicine. Like most medicines in the U.S., most of the cost of the new treatment will be paid by insurers, not patients, including private plans and government programs. After decades of research, gene therapies have begun reshaping the treatment of cancers and rare inheritable diseases with medicines that can modify or correct mutations embedded in people's genetic code. Hemgenix is the first such treatment for hemophilia and several other drugmakers are working on gene therapies for the more common form of the disorder, hemophilia A.

How Thinking Hard Makes the Brain Tired (economist.com) 47

An anonymous reader shares a report: Physical labour is exhausting. A long run or a hard day's sweat depletes the body's energy stores, resulting in a sense of fatigue. Mental labour can also be exhausting. Even resisting that last glistening chocolate-chip cookie after a long day at a consuming desk job is difficult. Cognitive control, the umbrella term encompassing mental exertion, self-control and willpower, also fades with effort. But unlike the mechanism of physical fatigue, the cause of cognitive fatigue has been poorly understood. Previous accounts were incomplete. One of the most widely known, the biological one, draws from what is known about muscular fatigue. It posits that exerting cognitive control uses up energy in the form of glucose. At the end of a day spent intensely cogitating, the brain is metaphorically running on fumes. The problem with this version of events is that the energy cost associated with thinking is minimal. One analysis of previous studies suggests that cognitively overworked and "depleted" brains use less than one-tenth of a Tic-Tac's worth of additional glucose.

If cognitive fatigue is not caused by a lack of energy, then what explains it? A team of scientists led by Antonius Wiehler of Pitie-Salpetriere University Hospital, in Paris, looked at things from what is termed a neurometabolic point of view. They hypothesise that cognitive fatigue results from an accumulation of a certain chemical in the region of the brain underpinning control. That substance, glutamate, is an excitatory neurotransmitter that abounds in the central nervous systems of mammals and plays a role in a multitude of activities, such as learning, memory and the sleep-wake cycle. In other words, cognitive work results in chemical changes in the brain, which present behaviourally as fatigue. This, therefore, is a signal to stop working in order to restore balance to the brain. In their new paper in Current Biology, the researchers describe an experiment they undertook to explain how all this happens.

Half in UK Back Genome Editing To Prevent Severe Diseases (theguardian.com) 120

More than half the UK backs the idea of rewriting the DNA of human embryos to prevent severe or life-threatening diseases, according to a survey. From a report: Commissioned by the Progress Educational Trust (PET), a fertility and genomics charity, the Ipsos poll found that 53% of people support the use of human genome editing to prevent children from developing serious conditions such as cystic fibrosis. There was less enthusiasm for use of the procedure to prevent milder conditions such as asthma, with only 36% in favour, and to create designer babies, with only a fifth expressing support, but views on the technology differed dramatically with age.

Younger generations were far more in favour of designer babies than older people, with 38% of 16- to 24-year-olds and 31% of 25- to 34-year-olds supporting the use of gene editing to allow parents to choose features such as their child's height and eye and hair colour. In the UK and many other countries it is illegal to perform genome editing on embryos that are intended for pregnancies, but the restrictions could be lifted if research shows the procedure can safely prevent severe diseases. Genome editing has been hailed as a potential gamechanger for dealing with a raft of heritable diseases ranging from cystic fibrosis and muscular dystrophy to Tay-Sachs, a rare condition that progressively destroys the nervous system. In principle, the faulty genes that cause the diseases can be rewritten in IVF embryos, allowing those embryos to develop into healthy babies.

Third Member of US FDA Advisory Panel Resigns Over Alzheimer's Drug Approval 61

A third member of a panel of outside advisers to the U.S. Food and Drug Administration has resigned in protest over the agency's decision to approve Biogen's Alzheimer's disease treatment despite the committee's recommendation against doing so. Reuters reports: Aaron Kesselheim, a Professor of Medicine at Harvard Medical School who had served on the FDA's advisory committee for nervous system drugs since 2015, told Reuters on Thursday he was stepping down from the panel. "My rationale was that the FDA needs to re-evaluate how it solicits and uses the advisory committees ... because I didn't think that the firm recommendations from the committee in this case ... were appropriately integrated into the decision-making process," Kesselheim said in an email. He cited FDA's decision to approve Sarepta Therapeutic's drug, eteplirsen, for Duchenne muscular dystrophy in 2016 as another example of the regulator approving a drug against the recommendations of its advisory committee.

On Tuesday, a member of the advisory group who voted against the approval, Washington University neurologist Dr. Joel Perlmutter, resigned from the committee, citing the FDA's approval of Aduhelm. Mayo Clinic neurologist Dr. David Knopman said he resigned on Wednesday. The 11-member committee voted nearly unanimously in November that Biogen's drug should not be approved, citing inconclusive evidence that the drug was effective.

EU Pushes for 'Right To Disconnect' From Work at Home (dw.com) 63

With ever more people working from home -- roughly a third of all employees within the bloc according to the Associated Press (AP) -- and needing to be constantly reachable, the boundaries between work and private life have become increasingly hazy. On Wednesday, EU lawmakers passed a non-binding resolution arguing that individuals have a fundamental "right to disconnect." The European Parliament Employment Committee voted 31-to-6, with 18 abstentions, in favor of allowing people to take time off and urged the European Commission to create rules that "catch up with the new reality" of work, according to Alex Agius Saliba, the Maltese Socialist politician who spearheaded the resolution.

"After months of teleworking, many workers are now suffering from negative side effects such as isolation, fatigue, depression, burnout, muscular or eye illnesses," said Saliba, adding: "The pressure to always be reachable, always available, is mounting, resulting in unpaid overtime and burnout." The committee measure must now be approved by the full chamber before it can be submitted to the Commission and EU member state governments for a vote. Lawmakers in favor of the resolution say the need for employees to be available via smartphone or e-mail around the clock is detrimental to mental health and well-being and that workers should be allowed to be offline without suffering employer retribution as a result.

As advertisers pull away from Facebook to protest the social networking giant's hands-off approach to misinformation and hate speech, the company is instituting a number of stronger policies to woo them back. From a report: In a livestreamed segment of the company's weekly all-hands meeting, CEO Mark Zuckerberg recapped some of the steps Facebook is already taking, and announced new measures to fight voter suppression and misinformation -- although they amount to things that other social media platforms like Twitter have already enacted and enforced in more aggressive ways.

At the heart of the policy changes is an admission that the company will continue to allow politicians and public figures to disseminate hate speech that does, in fact, violate the Facebook's own guidelines -- but it will add a label to denote they're remaining on the platform because of their "newsworthy" nature. It's a watered down version of the more muscular stance that Twitter has taken to limit the ability of its network to amplify hate speech or statements that incite violence. [...] Zuckerberg's remarks came days of advertisers -- most recently Unilever and Verizon -- announced that they're going to pull their money from Facebook as part the #StopHateforProfit campaign organized by civil rights groups.

Scientists Sent Mighty Mice To Space To Improve Treatments Back On Earth (npr.org) 12

In December, scientists sent 40 very muscular mice to live temporarily at the International Space Station. The resulting research, they hope, could lead to new treatments for kids with muscular dystrophy, or cancer patients with muscle wasting. From a report: In early December at the Kennedy Space Center in Florida, two anxious scientists were about to send 20 years of research into orbit. "I feel like our heart and soul is going up in that thing," Dr. Emily Germain-Lee told her husband, Dr. Se-Jin Lee, as they waited arm-in-arm for a SpaceX rocket to launch. A few seconds later the spacecraft took off, transporting some very unusual mice to the International Space Station, where they would spend more than a month in near zero gravity. Ordinarily, that would cause the animals' bones to weaken and their muscles to atrophy. But Lee and Germain-Lee, a power couple in the research world, were hoping that wouldn't happen with these mice.

"It was worth waiting 20 years for," Lee said as the Falcon 9 rocket headed toward space. "And someday it may really help people," Germain-Lee added. The couple hope that what they learn from these mice will lead to new treatments for millions of people with conditions that weaken muscles and bones. Among those who might eventually benefit: children with muscular dystrophy or brittle bone disease, cancer patients with muscle wasting, bedridden patients recovering from hip fractures, older people whose bones and muscles have become dangerously weak, and astronauts on long space voyages.

How Diet May Have Changed the Way Humans Speak (go.com) 45

"Ancient hunter-gathererers often had front teeth that met together, unlike today's more common alignment where the upper front teeth 'overbite' the lower front teeth," writes Slashdot reader omfglearntoplay. "This malocclusion is a result of changes to the ancestral human diet and introduction of soft foods, according to a new study published in the journal Science." ABC News reports: More than 2,000 different sounds exist across the roughly 7,000 to 8,000 languages that humans speak today, from ubiquitous cardinal vowels such as "a" and "i" to the rare click consonants found in southern Africa. Scientists had long thought this range of sounds was fixed in human biology since at least the emergence of our species about 300,000 years ago. However, in 1985, linguist Charles Hockett noted that labiodentals -- sounds produced by positioning the lower lip against the upper teeth, including "f" and "v" -- are overwhelmingly absent in languages whose speakers are hunter-gatherers. He suggested tough foods associated with such diets favored bites where teeth met edge on edge, and that people with such teeth would find it difficult to pronounce labiodentals, which are nowadays found in nearly half the world's languages.

To explore Hockett's idea further, researchers developed computer models of the human skull, teeth and jaw in overbite, overjet and edge-on-edge bite configurations. They next analyzed the amount of effort these configurations needed to pronounce certain labiodental sounds. The scientists found that overbites and overjets required 29 percent less muscular effort to produce labiodental sounds than edge-on-edge bites. In addition, overbites and overjets made it easier to accidentally mispronounce bilabial sounds such as "m," "w" or "p," which are made by placing the lips together, as labiodental ones.
The researchers also discovered that hunter-gatherer societies only have about 27 percent the number of labiodentals found in agricultural societies.

"Moreover, when they focused on the Indo-European language family -- which stretches from Iceland to the eastern Indian state of Assam and has records stretching back more than 2,500 years on how sounds in some of its languages were pronounced -- they found the use of labiodentals increased steadily following the development of agriculture," the report says. "All in all, they estimated that labiodentals only had a 3 percent chance of existing in the Indo-European proto-language that emerged about 6,000 to 8,000 years ago but are now found in 76 percent of the family's languages."

CRISPR Gene Editing Fixes Muscular Dystrophy In Dogs, Humans Could Be Next (time.com) 112

schwit1 shares a report from Time: In a new paper published in Science, researchers led by Eric Olson, professor and chair of molecular biology at UT Southwestern Medical Center, reported that he and his team successfully used CRISPR to correct the genetic defect responsible for Duchenne muscular dystrophy in four beagles bred with the disease-causing gene. It's the first use of CRISPR to treat muscular dystrophy in a large animal. (Previous studies had tested the technology on rodents.) In varying degrees, the genetic therapy halted the muscle degradation associated with the disease. Duchenne is caused by mutations in the dystrophin gene, which codes for a protein essential for normal muscle function. People born with the disease are often eventually confined to wheelchairs as their muscles continue to weaken, and in the later stages, many rely on ventilators to breathe as their diaphragm muscles stop working. Eventually, they develop heart and respiratory failure.

Olson and his team "fixed" the mutated dystrophin gene in four dogs by splicing out an offending section of the gene using CRISPR. The gene editing technology, discovered in 2012, can cut out sections of DNA at precise locations (and also potentially introduce new DNA as well). In the case of Duchenne, says Olson, simply snipping out a section of the mutated dystrophin gene allows the gene to make enough of the proper protein that muscles need to function. The hope is that if those animal studies and human trials prove this technique is safe and effective, CRISPR could potentially lead to a cure for Duchenne, Olson says. "We are going for a cure, not a treatment," he says. "All of the other therapies so far for Duchenne muscular dystrophy have treated the symptoms and consequences of the disease. This is going right at the root cause of the genetic mutation."

How the NSA Identified Satoshi Nakamoto (medium.com) 427

An anonymous reader shares a report: The 'creator' of Bitcoin, Satoshi Nakamoto, is the world's most elusive billionaire. Very few people outside of the Department of Homeland Security know Satoshi's real name. In fact, DHS will not publicly confirm that even THEY know the billionaire's identity. Satoshi has taken great care to keep his identity secret employing the latest encryption and obfuscation methods in his communications. Despite these efforts (according to my source at the DHS) Satoshi Nakamoto gave investigators the only tool they needed to find him -- his own words. Using stylometry one is able to compare texts to determine authorship of a particular work. Throughout the years Satoshi wrote thousands of posts and emails and most of which are publicly available. According to my source, the NSA was able to the use the 'writer invariant' method of stylometry to compare Satoshi's 'known' writings with trillions of writing samples from people across the globe. By taking Satoshi's texts and finding the 50 most common words, the NSA was able to break down his text into 5,000 word chunks and analyse each to find the frequency of those 50 words. This would result in a unique 50-number identifier for each chunk. The NSA then placed each of these numbers into a 50-dimensional space and flatten them into a plane using principal components analysis. The result is a 'fingerprint' for anything written by Satoshi that could easily be compared to any other writing. The NSA then took bulk emails and texts collected from their mass surveillance efforts. First through PRISM and then through MUSCULAR, the NSA was able to place trillions of writings from more than a billion people in the same plane as Satoshi's writings to find his true identity. The effort took less than a month and resulted in positive match.
