You just need to buy lot of 454 sequencing devices (http://www.454.com/) or Solexa http://www.solexa.com/wt/page/index and have big datacenter. Then you use those sequencers to re-sequence those 100 people and compare them with the reference human sequence. Big datacenter - You will need 30 Solexa devices and around 1000 TB data storage this is a nice task for Google datacenter.

I am Software Engineer in the Join Genome Institute at Lawrence Livermore National Laboratory. My research interests include distributed databases as well advanced visualization methods and their applications in bioinformatics. The second research interests area is in mobile solutions. In July 2002, the CarTooth Project Car Payment System, a group B.S. thesis, was awarded second place at the IEEE Computer Society's Second Annual International Design Competition, Washington DC. Currently I am developing Track Editor (Curration Tools) for Genome Portal at the JGI. At the LLNL, in 2005 I was helping develop techniques for the analysis of protein structure predictions within the 6th Community Wide Experiment on the Critical Assessment of Techniques (CASP) for the Protein Structure Prediction. The projects included design and implementation of the protein structure facility.
