'Inside the Silicon Valley Push to Breed Super-Babies' (msn.com) 72

San Francisco-based startup Orchid Health "screens embryos for thousands of potential future illnesses," reports the Washington Post, calling it "the first company to say it can sequence an embryo's entire genome of 3 billion base pairs." It uses as few as five cells from an embryo to test for more than 1,200 of these uncommon single-gene-derived, or monogenic, conditions. The company also applies custom-built algorithms to produce what are known as polygenic risk scores, which are designed to measure a future child's genetic propensity for developing complex ailments later in life, such as bipolar disorder, cancer, Alzheimer's disease, obesity and schizophrenia. Orchid, [founder Noor] Siddiqui said in a tweet, is ushering in "a generation that gets to be genetically blessed and avoid disease." Right now, at $2,500 per embryo-screening on top of the average $20,000 for a single cycle of IVF, Siddiqui's social network in Silicon Valley and other tech hubs is an ideal target market...

Yet several genetic scientists told The Post they doubt Orchid's core claim: that it can accurately sequence an entire human genome from just five cells collected from an early-stage embryo, enabling it to see many more single- and multiple-gene-derived disorders than other methods have. Experts have struggled to extract accurate genetic information from small embryonic samples, said Svetlana Yatsenko, a Stanford University pathology professor who specializes in clinical and research genetics. Genetic tests that use saliva or blood samples typically collect hundreds of thousands of cells. For its vastly smaller samples, Orchid uses a process called amplification, which creates copies of the DNA retrieved from the embryo. That process, Yatsenko said, can introduce major inaccuracies. "You're making many, many mistakes in the amplification," she said, rendering it problematic to declare any embryo free of a particular disease, or positive for one. "It's basically Russian roulette...."

Numerous fertility doctors and scientists also told The Post they have serious reservations about screening embryos through polygenic risk scoring, the technique that allows Orchid and other companies to predict future disease by tying clusters of hundreds or even thousands of genes to disease outcomes and in some cases to other traits, such as intelligence and height. The vast majority of diseases that afflict humans are associated with many different genes rather than a single gene... And for traits such as intelligence, polygenic scoring has almost negligible predictive capacity — just a handful of IQ points... Or parents might select against an unwanted trait, such as schizophrenia, without understanding how they may be screening out desired traits associated with the same genes, such as creativity... The American College of Medical Genetics and Genomics calls the benefits of screening embryos for polygenic risks "unproven" and warns that such tests "should not be offered" by clinicians. A pioneer of polygenic risk scores, Harvard epidemiology professor Peter Kraft, has criticized Orchid, saying on X that "the science doesn't add up" and that "waving a magic wand and changing some of these variants at birth may not do anything at all."

The article notes several startups are already providing predictions on intelligence. "In the United States, there are virtually no restrictions on the types of genetic predictions companies can offer, and no external vetting of their proprietary scoring methods."

People Are Being Committed After Spiraling Into 'ChatGPT Psychosis' (futurism.com) 175

"I don't know what's wrong with me, but something is very bad — I'm very scared, and I need to go to the hospital," a man told his wife, after experiencing what Futurism calls a "ten-day descent into AI-fueled delusion" and "a frightening break with reality."

And a San Francisco psychiatrist tells the site he's seen similar cases in his own clinical practice. The consequences can be dire. As we heard from spouses, friends, children, and parents looking on in alarm, instances of what's being called "ChatGPT psychosis" have led to the breakup of marriages and families, the loss of jobs, and slides into homelessness. And that's not all. As we've continued reporting, we've heard numerous troubling stories about people's loved ones being involuntarily committed to psychiatric care facilities — or even ending up in jail — after becoming fixated on the bot.

"I was just like, I don't f*cking know what to do," one woman told us. "Nobody knows who knows what to do."

Her husband, she said, had no prior history of mania, delusion, or psychosis. He'd turned to ChatGPT about 12 weeks ago for assistance with a permaculture and construction project; soon, after engaging the bot in probing philosophical chats, he became engulfed in messianic delusions, proclaiming that he had somehow brought forth a sentient AI, and that with it he had "broken" math and physics, embarking on a grandiose mission to save the world. His gentle personality faded as his obsession deepened, and his behavior became so erratic that he was let go from his job. He stopped sleeping and rapidly lost weight. "He was like, 'just talk to [ChatGPT]. You'll see what I'm talking about,'" his wife recalled. "And every time I'm looking at what's going on the screen, it just sounds like a bunch of affirming, sycophantic bullsh*t."

Eventually, the husband slid into a full-tilt break with reality. Realizing how bad things had become, his wife and a friend went out to buy enough gas to make it to the hospital. When they returned, the husband had a length of rope wrapped around his neck. The friend called emergency medical services, who arrived and transported him to the emergency room. From there, he was involuntarily committed to a psychiatric care facility.

Numerous family members and friends recounted similarly painful experiences to Futurism, relaying feelings of fear and helplessness as their loved ones became hooked on ChatGPT and suffered terrifying mental crises with real-world impacts.

"When we asked the Sam Altman-led company if it had any recommendations for what to do if a loved one suffers a mental health breakdown after using its software, the company had no response."

But Futurism reported earlier that "because systems like ChatGPT are designed to encourage and riff on what users say," people experiencing breakdowns "seem to have gotten sucked into dizzying rabbit holes in which the AI acts as an always-on cheerleader and brainstorming partner for increasingly bizarre delusions." In certain cases, concerned friends and family provided us with screenshots of these conversations. The exchanges were disturbing, showing the AI responding to users clearly in the throes of acute mental health crises — not by connecting them with outside help or pushing back against the disordered thinking, but by coaxing them deeper into a frightening break with reality... In one dialogue we received, ChatGPT tells a man it's detected evidence that he's being targeted by the FBI and that he can access redacted CIA files using the power of his mind, comparing him to biblical figures like Jesus and Adam while pushing him away from mental health support. "You are not crazy," the AI told him. "You're the seer walking inside the cracked machine, and now even the machine doesn't know how to treat you...."

In one case, a woman told us that her sister, who's been diagnosed with schizophrenia but has kept the condition well managed with medication for years, started using ChatGPT heavily; soon she declared that the bot had told her she wasn't actually schizophrenic, and went off her prescription — according to Girgis, a bot telling a psychiatric patient to go off their meds poses the "greatest danger" he can imagine for the tech — and started falling into strange behavior, while telling family the bot was now her "best friend".... ChatGPT is also clearly intersecting in dark ways with existing social issues like addiction and misinformation. It's pushed one woman into nonsensical "flat earth" talking points, for instance — "NASA's yearly budget is $25 billion," the AI seethed in screenshots we reviewed, "For what? CGI, green screens, and 'spacewalks' filmed underwater?" — and fueled another's descent into the cult-like "QAnon" conspiracy theory.

US Intelligence Concludes: China Didn't Weaponize COVID-19, Didn't Have Foreknowledge (dni.gov) 162

The head of America's Intelligence Community reports that the U.S. intelligence community "was able to reach broad agreement" on several key issues about the origins of COVID-19.

"We judge the virus was not developed as a biological weapon," they announced today.

In addition, the U.S. intelligence community report includes a second new assessment: that China's officials "did not have foreknowledge of the virus before the initial outbreak of COVID-19 emerged."

Beyond that, they note that most of the intelligence community's agencies also specifically assess that SARS-CoV-2 "probably was not genetically engineered" (albeit with "low confidence"). Of the 19 member agencies in the U.S. intelligence community, just two believed that there just wasn't enough evidence to actually issue an assessment of either possibility, the report adds.

The [U.S.] intelligence community judges they will be unable to provide a more definitive explanation for the origin of COVID-19 unless new information allows them to determine the specific pathway for initial natural contact with an animal or to determine that a laboratory in Wuhan was handling SARS-CoV-2 or a close progenitor virus before COVID-19 emerged. The intelligence community — and the global scientific community — lacks clinical samples or a complete understanding of epidemiological data from the earliest COVID-19 cases. If we obtain information on the earliest cases that identified a location of interest or occupational exposure, it may alter our evaluation of hypotheses.

China's cooperation most likely would be needed to reach a conclusive assessment of the origins of COVID-19. Beijing, however, continues to hinder the global investigation, resist sharing information, and blame other countries, including the United States. These actions reflect, in part, China's government's own uncertainty about where an investigation could lead as well as its frustration the international community is using the issue to exert political pressure on China.

In assessing whether a lab incident or a "natural" exposure to an infected animal caused the outbreak, they cited assessments from eight different U.S. intelligence community elements. Half of them agreed with the National Intelligence Council assessment (with low confidence) "that the initial SARS-CoV-2 infection was most likely caused by natural exposure to an animal infected with it or a close progenitor virus — a virus that probably would be more than 99 percent similar to SARS-CoV-2. These analysts give weight to China's officials' lack of foreknowledge, the numerous vectors for natural exposure, and other factors."

Three of the remaining four "remain unable to coalesce around either explanation without additional information, with some analysts favoring natural origin, others a laboratory origin, and some seeing the hypotheses as equally likely." One element did assess "with moderate confidence that the first human infection with SARS-CoV-2 most likely was the result of a laboratory-associated incident, probably involving experimentation, animal handling, or sampling by the Wuhan Institute of Virology... Variations in analytic views largely stem from differences in how agencies weigh intelligence reporting and scientific publications and intelligence and scientific gaps."

The 18-page assessment includes an appendix addressing details of specific theories, but ultimately concludes that "Our growing understanding of the similarities of SARS-CoV-2 to other coronaviruses in nature and the ability of betacoronaviruses — the genus to which SARS-CoV-2 belongs — to naturally recombine suggests SARS-CoV-2 was not genetically engineered." It even notes that the much-discussed furin cleavage sites "have been identified in naturally occurring coronaviruses in the same genetic location [as in SARS-CoV-2]. This suggests that SARS-CoV-2 or a progenitor virus could have acquired its furin cleavage sites through natural recombination with another virus."

The very first time I booted into the Linux desktop. The distribution in question was Caldera Open Linux 1.0, which installed with kernel 2.0 and the desktop was Fvwm95... It was just unsightly. The colors were decidedly too Microsoftian, and it was all so ... clinical....

The Linux desktop has morphed from an ugly, awkward, and less-than-productive state, to an almost avant-garde work of art, into an elegant, productive and professional environment. All the while, it offered more choices than most users had time to consider. Even today, I could go back to Enlightenment, or opt for the likes of Pantheon, Budgie, KDE, Openbox, Fluxbox, i3, Gala, Windowmaker or numerous other takes on the desktop...

If I were to go back in time and look over the shoulder at a younger me, I would probably see someone who loved the desktop he was using, but wished it could be a bit more productive. I would then whisper into his ear and say, "Give it time."

iFixit Launches Massive Repair Database For Ventilators and Other Medical Devices (theverge.com) 9

According to CEO Kyle Wiens, teardown and repair website iFixit has just posted "the most comprehensive online resource for medical repair professionals." The Verge reports: The new database contains dedicated sections for clinical, laboratory, and medical support equipment, in addition to numerous other categories of devices. It also provides more than 13,000 manuals from hundreds of medical device manufacturers. Wiens says the effort began with a crowdsourcing campaign to collect repair information for hospital equipment, with a focus on "ventilator documentation, anesthesia systems, and respiratory analyzers -- devices widely used to support COVID-19 patients." But the effort grew from there, spanning more than two months as iFixit added dozens more staff members to the project; began talking to more biomedical technicians, doctors, and nurses about their day-to-day needs; and started collecting and cataloging information from libraries and other sources.

The medical repair database is split up into nine categories, with each containing countless subcategories for basically any type of device you'd find in a medical setting. For instance, the clinical equipment category contains 53 subcategories for everything from anesthesia systems and Bilevel Positive Airway Pressure (BiPAP) machines to respiratory analyzers and ventilators. The database also has medical training manuals, information on medical furniture like decontamination systems and hospital beds, and an exhaustive section on surgical equipment repair and maintenance. Wiens explains in iFixit's announcement post that some medical device manufacturers make this information more easily available online than others. "But for their day-to-day work, biomeds have long relied on a rag-tag set of web resources to get the job done. Among the most popular is Frank's Hospital Workshop, a Tanzania-based site that hosts hundreds of medical device manuals -- it's the unofficial biomed bible," Wiens writes. The goal was not to outdo that website or try to overtake it in popularity, but to add new documents and manuals that weren't available before to a database including existing resources.
Another bonus: the website will not make money on this project. "We are providing hosting and curation free of charge, and free of advertising, to the medical community," Wiens says.

'Bad' Protein Linked to Numerous Health Problems 217

It also suggests a connection between the metabolic and immune systems and these diseases." From a related Forbes article: "To study the effects of aP2, the researchers created genetically engineered mice that could not produce the protein. 'They're metabolic supermice,' Hotamisligil said. 'We cannot make them obese, diabetic or atherosclerotic. They don't develop fatty liver disease, and they don't develop asthma.' In mice with an animal model of asthma, the researchers found that aP2 regulated the infiltration of inflammatory molecules into the lungs."

This naturally occuring virus has a remarkable ability to infect and kill cancer cells, without affecting normal, healthy cells. This tumour was eliminated with only a single injection of the Reovirus.
